以从陕西省泾阳县山羊大肠中采集的5条绵羊夏柏特线虫为研究对象,用通用引物JB3及JB4.5扩增绵羊夏柏特线虫的线粒体细胞色素c氧化酶第1亚基(cox1)基因部分序列(pcox1),将测序获得的序列用ClustalX 1.81程序进行比对,并用PAUP程序MP法构建系统发育树。结果表明:所获得的5个绵羊夏柏特线虫样品pcox1序列长度均为393bp;种系发育分析表明,绵羊夏柏特线虫5个样品位于同一分支,且绵羊夏柏特线虫的pcox1序列种内相对保守(0.5%~2.0%),种问变异显著(9.4%-18.8%),pcox1序列可作为种问变异研究的可靠遗传标记。
To analyze the sequence variation in the mitochondrial cytochrome c oxidase subunit 1 (cox1) gene among Chabertia ovina (C. ovina) isolates from goat in Shaanxi province, China and to study its phylogenetic relationships with other nematodes using the cox1 gene sequences, the partial cox1 (pcox1) was amplified from individual C. ovina samples by PCR. The amplified pcox1 sequences were aligned using the ClustalX 1.81 and MP tree was constructed using the software PAUP 4.0. The lengths of all the pcox1 sequences were 393 bp. Phylogenetic analyses showed that all the C. ovina samples from Shaanxi province were clustered in the same clade. Sequences variations in pcox1 with in C. ovina were limited (0.5%-2.0%), while inter-species differences were significant (9.4%-18.8%). Thus, the pcox1 sequences can be used as a genetic marker for identification and population genetic studies of nematodes.
