With the advent of new technologies, classifying human diseases on the basis of their specific gene expression patterns is becoming possible, such analyses are expected to yield diagnostic and prognostic cancer markers and to identify target molecules for anticancer drug development. For gene expression analysis, the presence of intact and extractable messenger RNA in the test material is mandatory. Archive tissue provide a affluent and huge resource library for these retrospective studies, but some studies were failed due to the content and integrity of nucleic acids was destroyed by tissue processing and fixative solution. So how to get high quality and more quantity nucleic acids is important for both clinicians and prospective investigators to be provide an overview of the methods of human tissue acquisition, fixation, and fixative condition, all of these is premise for successful studies.
