目的探究虫草素对肺癌细胞增殖的调节作用及其对细胞相应生理指标的影响。方法建立肺癌细胞体外培养体系,将生长状态良好的细胞进行分组,并分别与浓度为0、20、40、70、100、1 000μmol/L的虫草素共培养24h,通过MTT法测定肺癌细胞的增殖率,同时采用一氧化氮(NO)检测试剂盒测定各药物处理组细胞中NO的含量。结果20μmol/L及其以上浓度的虫草素能显著抑制肺癌细胞的增殖(P〈0.01),显著降低肺癌细胞中NO的相对含量(P〈0.05或0.01),且呈剂量依赖性。结论虫草素对肺癌细胞的增殖具有显著的抑制效果,且可能通过介导NO相关的信号途径抑制细胞增殖。
Objective To detect the role of cordycepin in adjusting the proliferation of lung carcinoma cell and affecting related physiological indicators of the cells. Methods The in vitro lung carcinoma cell culture system was constructed. The cells with growth in good condition were divided into groups, which were subject to co-cultured with cordycepin of different concentrations(0, 20, 40, 70, 100, 1 000 μmol/L) for 24 h, respectively. MTT method and NO kit was used for the detection of proliferation rate of lung carcinoma cell and NO content in the cells in each group, respectively. Results Cordycepin with a concentration more than 20 μmol/L could significantly inhibit the proliferation of lung carcinoma cells(P〈0.01), and significantly reduced NO production(P〈0.05 or 0.01). Conclusion Cordycepin shows active inhibitory effect on the proliferation of lung carcinoma cells, possibly through mediating relevant signal pathway of NO.
