目的:探讨祛瘀生肌法对糖尿病大鼠皮肤溃疡组织中基质金属蛋白酶-8(MMP-8)、基金金属蛋白酶-13(MMP-13)表达的影响效应。方法:以糖尿病大鼠皮肤溃疡模型为研究对象,采用不同配伍比例的生肌化瘀方进行干预,采用WesternBlot法和免疫组化法检测皮肤溃疡组织中MMP-8、MMP-13蛋白的表达。结果:WesternBlot法检测显示:生肌化瘀1、2、3号方组和正常创面组的MMP培蛋白表达均低于糖尿病溃疡创面组(P〈0.05);生肌化瘀2号方组的MMP-13蛋白表达高于糖尿病溃疡创面组和正常创面组(P〈0.05)。免疫组化法检测显示,生肌化瘀2、3号方组溃疡创面的MMP-8蛋白表达低于糖尿病溃疡创面组、正常创面组和生肌化瘀1号方组(P〈0.01,P〈0.05);生肌化瘀2号方组MMP-13蛋白表达高于正常创面组(P〈0.05)。结论:在创面修复的早期运用生肌化瘀3号方有利于促进创面愈合,在创面修复的后期运用生肌化瘀2号方有利于减少瘢痕组织的形成。
Objective: To study the effects of stasis-removing and muscle-regeneration therapy on the expression of MMP-8 and MMP-13 in skin ulcer tissue of diabetic rats. Methods: Different compatibility proportion Shengji Huayu Recipe (SHR) were used to treat the diabetic rats with skin ulcers. The expression of MMP-8 and MMP-13 protein in skin ulcers of the diabetic rats were detected by Western Blot method and immunohistochemical method. Results: Results of Western Blot showed that the MMP-8 protein expression of Shengji Huayu Recipe No.1 (SHR1) group, Shengji Huayu Recipe No.2 (SHR2) group, Shengxi Huayu Recipe No.3 (SHR3) group and normal ulcer group were lower than that of diabetic ulcer group (P〈0.05), and the MMP-13 protein expression of SHR2 group was higher than that of the diabetic ulcer group and normal ulcer group (P〈0.05). Immunohistochemistry results showed that MMP-8 protein expression of SHR2 group and SHR3 group were lower than that of diabetic ulcer group,normal ulcer group and SHR1 group (P〈0.01, P〈0.05) and the MMP-13 protein expression of SHR2 group was significantly higher than that of normal ulcer group (P〈0.05). Conclusion: Using SHR3 is conducive to the promotion of wound healing in the early wound repair stage, and using SHR2 is conducive to reducing the formation of scar tissue in later wound repair stage.
