①目的探讨血小板源性生长因子(PDGF)AKT通路在矽肺纤维化形成中的作用。②方法选用非暴露式气管灌注法复制大鼠矽肺模型,将其分为对照组(4周组和8周组)、矽肺模型组(4周组和8周组),采用免疫组织化学法检测磷酸化AKT、c-myc蛋白的定位与分布情况,免疫印迹法检测肺组织内PDGF、AKT、磷酸化AKT、c-myc以及I型和III型胶原蛋白的表达。③结果与相应的对照组相比,矽肺模型组大鼠肺组织内PDGF、磷酸化AKT、c-myc蛋白以及I型和III型胶原蛋白的表达均增加(P〈0.05);各组间比较AKT蛋白表达无明显改变。④结论 PDGF介导的AKT及其c-myc促进细胞增殖途径,促进了肺组织间质细胞的增生及其胶原的合成,促进了矽肺纤维化的形成与进展。
Objective To investigate whether activity of AKT pathway mediated by PDGF could play a promotion role in silicosis.Methods We use the method disposable endobrochial SiO2 powder douche to make the rat model with silicosis.Groups of experimental animals are as follows: control of silicotic model(control 4 and 8 weeks),silicotic model(control 4 and 8 weeks).Expressions of PDGF,phospho-AKT and AKT,c-myc and type I and type III collagen were detected by immunohistochemistry and western blot.Results Compared with the control group,expressions of PDGF,phosphor-AKT,c-myc and type I and type III collagen in silicotic model were all increased significantly(P0.05).While the expressions of AKT have no significant difference among each groups.Conclusion The activity of AKT/c-myc pathway mediated by PDGF possibly induces pulmonary mesenchymal proliferation and collagen synthesis in rats,which promotes forming and development of silicosis.
