以156条随机引物,对目前中国落叶松杨栅锈菌(Melampsora larici-populina Kleb)的主要优势菌系进行了RAPD片段的筛选,寻找到了1号3、号4、号5、号4个流行生理小种的特异性RAPD标记。经过多批次扩增,引物S205和S284分别得到1号生理小种长度约450和760 bp的特异条带,S25得到3号生理小种长度约1440 bp的特异条带,S51和S286得到4号生理小种长度约970和1020 bp的特异条带,S23和S96分别得到5号生理小种长度约750和780 bp的特异条带。此结果表明,通过寻找落叶松杨栅锈菌生理小种的特异性RAPD片段,能够建立起中国落叶松杨栅锈菌生理小种的分子检测体系。
156 random primers was used to screen main dominance fungus strain of Chiese Melampsora larici-populirta Kleb by RAPD technique and the special RAPD fragments of physiological races of C1, C3, CA and C5 were founded successfully. Through many batches of amplification, the primers of S205 and S284 obtained C1 special fragment with race length about 450 and 760 bp respectively;S25 obtained C3 special fragment with race length about 1 440 bp;SSland S286 obtained CA special fragment with race length about 970 and 1 020 bp respectively and S23 and S96obtained C5 special fragment with race length about 750 and 780 bp respectively. the results indicated that a molecular identification system for the physiological races of Melampsora larici-populina in China could be established through cosmically searching special RAPD fragments of different races.
