对利用cDNA—AFLP技术获得的“紫娟”茶树幼嫩叶与成熟叶之间的差异表达片段TDF,通过RACE方法首次从茶树中克隆了羟甲基戊二酰辅酶A合酶基因的全长cDNA,命名为CsHMGS(GenBank登录号为JQ390224)。CsHMGS全长1829bp,开放阅读框1401bp,编码467个氨基酸,经氨基酸序列对比发现,CsHMGS编码的氨基酸序列与人参、橡胶树、春花和喜树的HMGS基因同源性分别为87%、86%、87%和87%,含有HMGS蛋白保守序列,表达特性分析发现在成熟叶片中表达量高于幼嫩叶片。
A TDF has been gained from genes expressed differentially between young and mature leav es of tea plant(Camellia sinensis var. assamica, cultivar Zijuan)using cDNA-AFLP. In this study, a 3-hydroxy 3-methylglutaryl-CoA synthase (HMGS)gene was cloned from tea plant by RACE. The full-length HMGS cDNA of tea plant (designated as CsHMGS, GenBank accession No. JQ390224) was 1 829 bp and contained a complete open reading frame (ORF) of 1 401 bp, which encoded 467 amino acid residues. It was found that the amino acid sequence of CsHMGS had more than 85% homology with Panax ginseng, Hevea brasiliensis, Catharanthus roseus, Camptotheca acuminate, and the conserved sequence of HMGS protein was also found in CsHMGS. Expression analysis found that CsHMGS was higher in mature leaves than in young ones.
