目的采用高压液相色谱法(HPLC)测定"黄芩-黄连"药对中黄芩4种有效成分(黄芩苷、汉黄芩苷、黄芩素、汉黄芩素)。方法采用Hypersil ODS2-C18色谱柱(250 mm×4.6 mm,5μm);流动相为乙腈-0.3%磷酸三乙胺,梯度洗脱,流速:1.0 mL·min^(-1),检测波长:280 nm,柱温:30℃。结果黄芩苷、汉黄芩苷、黄芩素和汉黄芩素分别在0.048-0.288 mg·mL^(-1)、0.056-0.336 mg·mL^(-1)、0.042-0.254 mg·mL^(-1)和0.047-0.280mg·mL^(-1)范围线性关系良好,平均回收率分别为100.01%、99.99%、99.64%、99.79%,RSD分别为1.20%、0.80%、1.04%、0.94%。结论该方法简便,结果准确可靠,可作为"黄芩-黄连"药对提取物的质量控制标准。
Objective To establish a HPLC method for the determination of 4 constituents(baicalin, wogonoside,baicalein, and wogonin) from extract of herb pair of Radix Scutellariae and Rhizoma Coptidis. Methods The determination was analyzed on Hypersil ODS2- C18 column with acetonitrile- 0.3 % phosphoric acid- triethylamine as mobile phase at gradient elution program. The flow rate was 1.0 mL·min^-1,the detection wavelength was set at 280 nm,and column temperature was 30 ℃. Results A good linearity was shown in the range of 0.048- 0.288 mg·mL^-1 for baicalin, 0.056- 0.336 mg·mL^-1 for wogonoside, 0.042- 0.254 mg·mL^-1 for baicalein, and 0.0466- 0.2795mg·mL^-1 for wogonin. The average recovery was 100.01 %, 99.99 %, 99.64 %, 99.79 % with relative standard deviation of 1.20 %, 0.80 %, 1.04 %, 0.94 % for baicalin, wogonoside, baicalein, wogonin, respectively.Conclusion The developed method provides a scientific basis for the quality assessment of the extracts of Radix Scutellariae and Rhizoma Coptidis herb pair.
