目的:探索江枳壳润肠颗粒治疗功能性便秘的药效学研究。方法:大鼠72只,分为正常对照组、模型对照组、聚乙二醇散剂(阳性对照组,2.08g/kg)和枳壳润肠颗粒5.5mg/kg、11mg/kg、22mg/kg组,除正常对照组外,其余各组灌胃50mg/kg复方地芬诺酯混悬液复制大鼠功能性便秘模型,记录首粒粪便排出时间及粪便含水率,观察对肠道推进运动的影响,ELISA法测定各组大鼠血清和结肠组织中的胃动素(MTL)、血管活性肠肽(VIP)、P物质(SP)的含量。结果:枳壳润肠颗粒5.5mg/kg、11mg/kg、22mg/kg能缩短大鼠首粒粪便排出时间,增加粪便含水率,提高大鼠肠道推进率,提高大鼠血清和肠道SP、MTL水平,降低血清VIP水平。结论:枳壳润肠颗粒6mg/kg、12mg/kg、24mg/kg对功能性便秘均有显著改善作用,其改善作用与促进胃肠运动,提高胃肠道的收缩力和张力有关。
Objective: To explore pharmacodynamic study of Citrus aurantium L.particles in treating functional constipation.Methods: 72 rats were divided into normal group,model group,polyethylene glycol powder( positive control group,2.08g/kg) and Citrus aurantium L.particles 5.5mg/kg,11mg/kg,22mg/kg.In addition to the normal group,the other groups were given 50mg/kg diphenoxylate suspension to induce functional constipation,and the time of the first grain feces fecal and water content were recorded and the effect of Citrus aurantium L.particles on intestinal propulsion movement was observed,ELISA method was used in detecting the content of serum and colon tissue of motilin( MTL),vasoactive intestinal peptide( VIP),P substance( SP) in rats.Results: Citrus aurantium L.particles 5.5mg/kg,11mg/kg,22mg/kg in rats can shorten the time of the first grain feces,increase stool water content,improve intestinal propulsion rate,increase serum and intestinal SP,MTL level,decrease serum VIP levels in rats.Conclusion: Citrus aurantium L.particles 6mg/kg,12mg/kg,24mg/kg can significantly improve functional constipation,improve and promote gastrointestinal motility,improve gastrointestinal contractility and tension related.
