为探讨青钱柳多糖对高脂血症小鼠脂肪酸合成酶基因表达的影响,通过建立高血脂模型,每天以不同剂量的多糖(100、200和400 mg/kg体重)进行灌胃,以辛伐他汀(4 mg/kg体重)做为阳性对照,持续喂养四周,采用半定量PCR和Western Blot检测小鼠肝脏和脂肪组织FAS mRNA和蛋白表达水平。结果表明,青钱柳多糖能抑制小鼠肝脏组织中FAS mRNA表达水平(P〉0.05),而对脂肪组织中FAS mRNA表达水平影响不明显;青钱柳多糖能显著抑制小鼠肝脏组织中FAS蛋白表达水平,以高剂量的多糖效果最佳,与模型组相比,差异极显著(P〈0.01);同时也能轻微抑制脂肪组织中FAS蛋白表达水平。结果提示,青钱柳多糖对高脂血症小鼠各内脏中FAS mRNA和FAS蛋白表达具有一定的影响作用。
To investigate the effect of Cyclocarya paliurus (Batal) Iljinskaja polysaccharide on the fatty acid synthase (FAS) gene and protein expression in hyperglecimia mice, the hyperglecimia mice model was in- duced with high-fat emulsion, then the mice were fed with different doses of polysaccharide (100,200,400 mg/kg body weight), and the positive control was fed with simvastatin (4 mg/kg body weight). After four weeks of feeding, the FAS mRNA and protein expressions of liver and adipose were detected by RT-PCR and Western Blot. The results showed that the polysaccharide could decrease the expression of FAS mRNA in the liver (P 〉 0.05 ), while the effect of the polysaccharide on adipose tissue was not obvious. Furthermore, the polysaccharide could significantly decrease the protein expression in the liver, and the effect of the high dosage polysaccharide was the best ( P 〈 0.01 ) compared with the model group, also the polysaccharide could slightly restrain the expression of FAS protein in the adipose. The results indicated that the effects of Cyclocarya pali- urus (Batal) Iljinskaja polysaccharide on the expressions of FAS mRNA and FAS protein in different tissues were different.
