目的:观察养心通脉有效部位方(apr-YTF)对大鼠骨髓间充质干细胞(BMSCs)归巢梗死心肌的影响。方法:将急性心肌梗死大鼠模型随机分成apr-YTF注射液、rhG-CSF注射液和PBS缓冲液3组,测量各组大鼠干预后在体心功能、心室肌瘢痕面积、瘢痕毛细血管数、心肌CD34+细胞、Brdu与cTn I双染阳性细胞数、Ⅷ因子、VEGF、bFGF、Flk-1、外周血CD34+细胞数,分析组间差异。结果:①与PBS组比较,apr-YTF组和rhG-CSF组的LVSP、LVEDP、+dp/dtmax、-dp/dtmax均有显著改善,心室肌瘢痕面积显著减小,心室肌毛细血管数、心肌边缘区和外周血液CD34+染色阳性细胞数、心肌梗死边缘区中Brdu与cTn I双染阳性细胞数及Ⅷ因子、Flk-1、VEGF、bFGF的表达均显著增加(P〈0.01,P〈0.05);②与rhG-CSF组比较,apr-YTF组的LVSP显著升高,心肌组织CD34+染色阳性细胞数、心肌梗死边缘区中Brdu与cTn I双染阳性细胞数均显著增高(P〈0.01)。结论:apr-YTF能促使BMSCs动员入血、定向归巢并转化为心肌细胞,且优于rhG-CSF。
Objective: To observe the effect of Yangxin Tongmai Formula effective components recipe on bone marrow mesenchymal stem cells homing to the infarcted myocardium in rats. Methods: The acute myocardial infarction (AMI) rat model was devided into apr-YTF injection group, rhG-CSF injection group, PBS buffer solution group, then the rat heart function in vivo was examined; the ventricular scar area and capillary counts were detected; the CD34+ and myocardial Brdu and cTnI double dyeing positive cells counts, and the content of factor VHI, VEGF, bFGF, Flk-1 in the myocardial tissue were detected; the peripheral blood CD34+ cell counts was also detected, the differece between the three groups was analyzed. Results: (.])Compared with PBS group, the LVSP, LVEDP, +dp/dtmax, -dp/dtmax were significantly improved in apr-YTF group and rhG-CSF group, the ventricular scar area percentage was decreased, the ventricular myocardial capillary counts, the CD34+ cell counts in the myocardial infarction marginal region and peripheral blood, Brdu and cTn I double dyeing positive cells counts in the myocardial infarction marginal region, the expression of factor VII, Flk-1, VEGF, bFGF in the myocardial infarction marginal region in apr- YTF group and rhG-CSF group were significantly increased (P〈0.01, P〈0.05 ). (~)Compared with rhG-CSF group, moreover, the LVSP, and the myocardial tissue CD34+ cell counts, myocardium, and the Brdu and cTnI double dyeing positive cells counts in apr-YTF group was significantly increased (P 〈 0.01). Conclusion: apr-YTF can promote BMSCs migrating to the blood, oriented homing and transferring to be mycardiocytes, which is better than rhG-CSF.
