目的:观察正常大鼠触液核内BKca-α分布及其在神经病理性疼痛过程中的表达变化,探讨触液核在神经病理性疼痛中的作用。方法:采用霍乱毒素B结合辣根过氧化酶(CB-HRP)和BKca-α免疫荧光双重标记技术,观测大鼠触液核中BKca-α在正常及慢性坐骨神经压迫损伤(chronic constriction injury,CCI)大鼠的表达变化。结果:正常和CCI大鼠触液核内存在BKca-α;在CCI大鼠,大鼠热痛觉及机械痛觉阈值下降,且触液核内BKca-α表达的数目也相应减少,和对照组比较有显著差异。结论:触液核BKca-α的降低可能参与神经病理性疼痛。
Objective : To observe the expression of BKca-α in CSF-contacting nucleus(CSF-CN) of normal rats and changes in it's expression during the process of neuropathic pain,and to discuss the role of BKca-αin CSF-CN in neuropathic pain.Methods: The CB-HRP/BKca-α immunofluorescence double-labeling method was used to observe the expression of BKca-α in CSF-CN in normal and chronic constriction injury(CCI) rats.Results: BKca-α was found in CSF-CN in normal and CCI rats.The thresholds of thermal and mechanical pain were remarkably decreased in CCI rats.Compared with sham rats,the expression of BKca-α in CSF-CN was decreased in CCI rats.Conclusion: The decrease of BKca-α in CSF-CN may play an important role in neuropathic pain.
