目的研究枇杷叶三萜酸(TAL)对大鼠肺纤维化的防治作用并探讨其对脂质过氧化的作用。方法通过气管内一次性滴注盐酸平阳霉素(5mg/kg)复制大鼠肺纤维化模型。治疗组于造模后第2天开始灌胃给予不同剂量的TAL或醋酸泼尼松。各组大鼠于给药后第7、14、28天随机取样6只,取固定部位肺组织石蜡切片HE染色,并分别观察各组大鼠肺脏指数、肺组织羟脯氨酸(HYP)、血清丙二醛(MDA)的含量和超氧化物歧化酶(SOD)的活力。结果病理学切片显示TAL能明显改善模型组大鼠肺脏组织结构,减轻肺纤维化增生程度。同时,TAL能明显降低模型组大鼠肺脏指数和肺组织中HYP的含量,并可提高大鼠血清中SOD的活力,降低血清中MDA的含量。结论TAL对大鼠肺纤维化有一定的防治作用,其机制可能与抗脂质过氧化有关。
Objective To investigate the therapeutic effects of triterpene acids of loquat(TAL)on Bleomycin A5-induced pulmonary fibrosis of rats and to explore the role of TAL on lipid peroxidation.Methods The model of pulmonary fibrosis was induced by injecting with a single dose of Bleomycin-A5(5 mg/kg)by trachea.From the second day,rats in therapeutic groups were treated with the related dose of TAL or prednisone.On the 7 th,14 th and 28 th day after medication,the levels of HYP in lung tissue,MDA and SOD levels in serum were measured.HE staining was used to evaluate the degree of pulmonary fibrosis.Results Lung histopathological examination shows TAL could ameliorate lung formation and alleviate fibrogenesis.At the same time,TAL not only reduced the lung index and HYP in lung tissue significantly,but also enhanced SOD activity and reduced MDA contents in serum obviously.Conclusion TAL has positively therapeutic effects on lung fibrosis,which may be accociated with its anti-lipid peroxidation effect.
