转录中介因子2(transcriptional intermediary factor 2,TIF2)也称糖皮质激素受体相互作用蛋白质1(glucocorticoid receptor interacting protein 1,GRIP1),作为类固醇受体辅激活蛋白(SRC)/p160家族(steroid receptor co-activator/p160 family)成员之一,通过与其他的核受体超家族成员相互作用发挥其功能。它是一种蛋白质共活化因子,可以辅助多种转录因子进而在转录水平上调多种基因的表达。上游的配体、信号通路和转录因子类型决定TIF2的作用,并在不同的组织细胞中表现出一定的特异性及复杂性。而对TIF2的研究有利于更深入地了解机体相关基因转录的精细调节和相关性疾病的可能机制,并为分子靶向治疗提供新的靶点。本文从TIF2蛋白的结构、染色质修饰及其在相关疾病中的作用等方面对近年来的最新进展予以综述。
Transcriptional intermediary factor 2 is one member of the p160 steroid receptor co-activator(SRC) gene family,which interacts with nuclear receptors.It serves as transcriptional co-activator of various transcription factors regulating a variety of gene expression.The effect of TIF2 is determined by the upstream ligands,signaling pathway and transcription factors.TIF2 shows certain specificity and complexity in different tissues and cells.So further studies on TIF2 will help us to understand transcriptional regulation of the related genes and possible mechanism of the related diseases,and provide a new approach for molecular targeted therapy.Here the paper reviews the progress in the fields of TIF2 protein structure,chromatin modification and TIF2-related diseases.
