目的探讨甲醛对小鼠体细胞染色体端粒的损伤。方法对昆明小鼠灌胃不同剂量的甲醛(10、20、40mg/kg)连续7d,通过PCR方法检测小鼠血细胞、肝细胞、肾细胞染色体端粒DNA重复序列的相对长度(0D260)。结果与对照组比较,10、20和40mg/kg组小鼠的血细胞、肝细胞和肾细胞染色体端粒DNA重复序列缩短,差异有统计学意义(P〈0.05,P〈0.01),且随着甲醛染毒剂量的升高呈缩短趋势。结论甲醛暴露能破坏小鼠染色体端粒的稳定。
Objective In order to explore the damage mechanism of chromosome telomere in mice indnced by formaldehyde. Methods Kunming mice were exposed to formaldehyde at different doses (10, 20, 40 mg/kg)through gavage for seven consecutive days, then chromosome telomere DNA repeat sequence relative length (OD260)of all mice's blood, liver and kidney cells were detected by PCR method in different groups. Results The length of chromosome telomere DNA repeat sequence of blood, liver and kidney cells shortened in 10, 20 and 40 mg/kg formaldehyde groups, moreover, with the increase in exposure doses, telomere DNA length (OD260) had statistical significance compared with the control group. Conclusion Formaldehyde can damage chromosome telomere stability in tissue cells.
