目的探讨姜黄素联合细胞因子活化的杀伤细胞(CIK cells)杀伤肾癌细胞逆转耐药P-糖蛋白(P-gp)表达的作用。方法培养耐阿霉素的肾癌细胞系(786-O/ADR),观察不同浓度姜黄素和CIK细胞单独或联合处理后,应用cck-8检测细胞活性,免疫荧光检测P-gp的表达变化,TUNEL和caspase-3染色观察细胞凋亡情况。结果姜黄素(浓度分别为20μM和40μM)、CIK细胞(分别为20:1和40:1)单独应用以及姜黄素+CIK细胞联合应用组均能显著降低P-gp的表达水平,降低细胞活性,下调caspase-3表达水平(〈0.05)。结论姜黄素联合CIK细胞逆转肾癌细胞耐药蛋白P-gp表达,促进细胞凋亡,发挥协同作用。
Objective To investigate the effect of combination of curcumin and cytokine-induced killer(CIK) cells on the expression of P-glucose protein in renal carcinoma786-O/ADR cells. Methods The renal carcinoma cell Doxorubicin-resistance cell line(786-O/ADR) was cultured and treated by different concentrations of curcumin and CIK cells individually or in combination. The cell viability was observed by CCK-8 kit, the expression of P-gp was observed by immunofluorescence, and the apoptosis was observed by caspase-3 immunostaining and TUNEL assay. Results The expression levels of P-gp in 786-O/ADR cells and the cell viability were decreased significantly( 〈0.05), and the expression level of caspase-3 was downregulated after treatment of curcumin(20μM and 40μM) or CIK cells(20 : 1 and 40 : 1) or the combination of curcumin+ CIK cells. Conclusion The combination of curcumin and CIK cells reverses the expression of P-glucose protein in 786-O/ADR cells, would be used in treatment of renal carcinoma.
