目的观察白花蛇舌草总黄酮(FOD)对体外培养黑色素瘤A375和B16细胞增殖的影响。方法体外培养黑色素瘤A375和B16细胞,分别使用浓度为6.25、12.5、25、50、100 mg.L-1的FOD作用24、48 h,通过MTT比色法检测细胞生长的抑制作用。结果 12.5 mg.L-1的FOD作用A375细胞48 h后,细胞增殖能力明显受抑制(P〈0.05),25 mg.L-1的FOD作用A375细胞24 h后,细胞增殖能力也明显受抑制(P〈0.05);25 mg.L-1的FOD作用B16细胞48 h后,细胞增殖活力明显下降(P〈0.05),50 mg.L-1的FOD作用B16细胞24 h后,细胞增殖活力也明显降低(P〈0.05);分别作用24 h和48 h后,两组随着浓度的升高,不同浓度对细胞的增殖能力抑制差异有统计学意义(P〈0.05);随着浓度的增高和作用时间的延长,细胞的增殖能力随着下降。结论 FOD能有效抑制黑色素瘤A375和B16细胞增殖,且具有一定的时-效和量-效关系。
Objective To observe the effect of the total flavones of oldenlandia diffusa wild(FOD)on the proliferation of melanoma A375 and B16 cells.Methods The melanoma A375 and B16 cells were cultured in vitro with the concentration of 6.25 mg·L-1,12.5 mg·L-1,25 mg·L-1,50 mg·L-1 and 100 mg·L-1 of FOD for 24 h and 48 h separately.MTT assay was adopted to detect the cell growth inhibition.Results With FOD 12.5 mg·L-1 for 48 h,the proliferation of A375 was inhibited apparently(P0.05).With FOD 25 mg·L-1 for 24 h,the proliferation of A375 was inhibited obviously,too(P0.05).With FOD 25 mg·L-1 for 48 h,the activity of B16 proliferation decreased apparently(P0.05).With FOD 50 mg·L-1 for 24 h,the activity of B16 proliferation decreased apparently,too(P0.05).Separately,after 24 h and 48 h,with the concentration increasing of FOD,the statistical difference presented in the inhibition on the cell proliferation(P0.05).While increasing the concentration and culturing for longer time,the cell proliferation was reducing.Conclusion FOD effectively inhibits the proliferation of melanoma A375 and B16 cells,and indicates a certain time-efficiency and capacity-effect relationship.
