目的评价脊髓胶质细胞在树酯毒素(RTX)诱导的神经病理性疼痛大鼠的作用。方法雄性健康SD大鼠60只随机均分为两组:RTX组单次腹腔注射RTX 210μg/kg建立神经病理性疼痛模型;C组注射等量溶媒(10%Tween 80、10%乙醇与生理盐水组成的混合液)作为对照。检测两组大鼠建模前及建模后1、3、7、42d机械痛阈值(PWMT)及热痛阈值(PWTL)。分别采用qRT-PCR和Western blot在建模后4h及1、3、7、42d检测CD11b及胶质纤维酸性蛋白(GFAP)mRNA和蛋白表达水平。结果与建模前相比,RTX组建模后3、7、42d大鼠PWMT下降和PWTL升高(P〈0.05)。与C组相比,RTX组CD11bmRNA及蛋白表达水平在建模后4h升高(P〈0.05),GFAP mRNA及蛋白表达水平在建模后4h及1、3、7、42d升高(P〈0.05)。RTX组GFAP表达与PWMT呈负相关(r=-0.96,P〈0.01)。结论激活的神经胶质细胞可能在神经病理性疼痛的产生与维持阶段起重要作用。
Objective To evaluate the effect of spinal glial cells in rats with resiniferatoxin(RTX)-induced neuropathic pain.Methods Sixty male SD rats were equally divided into two groups.The rats in group RTX were induced by a single intraperitoneal injection of RTX 210μg/kg to build neuropathic pain models.The rats in group C were treated with intraperitoneal injection of equivalent mixture of 10% Tween 80,10% alcohol and normal saline.Paw withdrawl mechanical threshold(PWMT)and paw withdrawal thermal latency(PWTL)in two groups were examined before and on the 1st,3rd,7th and 42nd day after modeling.The expressions of CD11 band glial fibrillary acidic protein(GFAP)mRNA and their proteins were detected at 4 hours and on the 1st,3rd,7th,42nd day after modeling by qRT-PCR and Western blot,respectively.Results Compared with before,PWMT was decreased and PWTL was increased on the 3rd,7th and 42nd day after model establishment in group RTX(P〈0.05).Compared with group C,the levels of CD11 bmRNA and protein were increased at 4 hours and after modeling,levels of GFAP mRNA and protein were increased at 4 hours and on the 1st,3rd,7th,42nd day after model establishment in group RTX(P〈0.05).GFAP expression was negatively correlated to PWMT(r=-0.96,P〈0.01).Conclusion Activated glial cells may play an important role in the formation and maintenance of neuropathic pain.
