为获得抵抗逆境胁迫的相关基因,利用电子克隆方法从水稻中获得了1个新基因,将其命名为OsKAT2(GenBank登陆号为KJ010541)。生物信息学分析结果表明OsKAT2的cDNA序列开放阅读框长1806 bp,编码蛋白质由601个氨基酸组成;OsKAT2蛋白质包含K+通道的保守结构域,属于Shaker家族第II组。通过实时定量荧光PCR分析发现,OsKAT2主要在地上部表达;在干旱、高盐和外源脱落酸( ABA)胁迫处理下,水稻地上部OsKAT2的表达量均降低。初步推测OsKAT2可能在水稻抵抗逆境胁迫中发挥作用,是一个水稻抗非生物逆境改良的候选基因。
A new gene, designated OsKAT2 (GenBank accession number KJ010541), was isolated from Orazy sati-va by in silico cloning. Bioinformatics analysis showed that the open reading frame of OsKAT2 was 1 806 bp in length enco-ding 601 amid acid residues. The protein OsKAT2 which contained K+ channel conserved domains belonged to group II of family Shaker. Real-time fluorescence quantitative PCR ( RT-qPCR) revealed OsKAT2 was localized in shoot. The expres-sion levels of OsKAT2 declined under drought, salt and in vitro ABA stress. The results indicated that OsKAT2 might play a role in abiotic-stress-tolerance, and could be a candidate gene for rice abiotic-stress-tolerance improvement.
