目的探讨狼毒大戟石油醚提取物对小鼠肝脏细胞凋亡及磷酸化细胞外信号调节激酶(p-ERK)、Bcl-2、Bax、半胱氨酸蛋白酶(Caspase)-3表达的影响。方法 80只昆明种小白鼠随机分为低剂量组、中剂量组、高剂量组和植物油对照组,每组20只。采用腹腔注射法给药,1次/d(0.39、0.78、1.55 mg·kg^-1·d^-1)。于实验第14天分别进行体重测量,对小鼠肝脏组织进行HE染色,TUNEL法原位标记凋亡细胞,采用免疫组织化学染色检测Bcl-2、Bax蛋白表达,Western印迹法检测p-ERK、Caspase-3蛋白表达。实时定量PCR对Bcl-2、Bax、Caspase-3进行定量分析。结果与植物油对照组比较,高剂量组小鼠体重明显下降,肝脏细胞发生明显凋亡,p-ERK、Bcl-2表达下调,Bax表达上调(P〈0.05),Caspase-3产生活化形式的Cleaved-Caspase-3。结论狼毒大戟石油醚提取物可使小鼠肝脏细胞发生凋亡,其机制可能与降低p-ERK、Bcl-2,增强Bax蛋白表达,并使Caspase-3产生活化形式的Cleaved-Caspase-3有关。
Objective To investigate the effect of petroleum ether extracts of Euphorbia fischeriana on the liver cells apoptosis in mice and the expressions of phosphorylation of extracellular signal-regulated kinase( p-ERK),Bcl-2,Bax,Caspase-3 in the liver of mice.Methods Eighty Kunming mice were randomly divided into low dose,middle dose,high dose and the control groups with vegetable oil( 0. 39,0. 78,1. 55 mg·kg^-1·d^-1),20 in each group. Model was made by intraperitoneal injection 1 time/day. In the experiment of 14 days,weight was measured,HE dyeing was used to detect liver pathologic morphology,TUNEL was used to detect liver cell apoptosis. Bcl-2 and Bax protein expression were tested by immunohistochemical staining method. The protein expressions of p-ERK and Caspase-3 were detected by western blot. Real-time PCR was used for quantitative analysis of Bcl-2,Bax and Caspase-3. Results Compared with the control group,the weights were decreased significantly,liver cell apoptosis was more obvious,the protein expressions of p-ERK and Bcl-2 were decreased while Bax were increased,the protein expressions of Caspase-3 was activated to Cleaved-Caspase-3 in high dose group( P〈0. 05).Conclusions Petroleum ether extracts of Euphorbia fischeriana could result in apoptosis of liver cells in mice. The mechanism may be related to reduce p-ERK and Bcl-2,enhance Bax protein expression,and produce activated Caspase-3 forms of Cleaved-Caspase-3.
