以‘津研四号’黄瓜品种为材料,克隆得到一个黄瓜膜联蛋白基因的cDNA全长,命名为CsANN2。为进一步探究黄瓜CsANN2基因的功能,对CsANN2基因编码的蛋白序列进行生物信息学分析后,发现其理化性质、保守结构域以及多个功能蛋白位点都和其他植物的膜联蛋白一致。系统进化树分析表明,黄瓜CsANN2与AtANN2、AtANN6、AtANN7、BjANN2和BjANN6有较近的亲缘关系。亚细胞定位结果显示,CsANN2蛋白在细胞膜、细胞质与细胞核内都有分布。qRT-PCR结果分析表明,CsANN2基因受ABA、SA、PEG和H2O2诱导表达,但对NaCl胁迫无响应。
A full length cDNA,designated as CsANN2,was isolated from the cucumber 'Jinyan 4' (Cucumis sativus L. ). Bioinformatics analysis of the deduced CsANN2 protein revealed that its physiochemicaJ properties, conserved domain and multifunction site were consistent with other ANNs of other plants. The phylogenetic tree indicated that CsANN2 had closer relationship with AtANN2, AtANN6, AtANN7, BjANN2, BjANN6. The subcellular localization assay indicated that CsANN2 located in membrane,cytoplasm and nucleus. The quantitative RT-PCR results showed that the expression of CsANN2 was induced by ABA,SA, PEG and H2O2, but not affected by NaCI. The current study formed a base for probing the biological functional of CsANN2 in cucumber.
