目的:确定阿维A对寻常型银屑病表皮中早幼粒白细胞基因调节的影响。方法:用原位杂交和免疫组化法检测进行期寻常型银屑病皮损阿维A治疗前后早幼粒细胞白血病(PML)基因mRNA及蛋白的表达。结果:阿维A治疗前早幼粒白细胞基因和蛋白在寻常型银屑病皮损中均有高表达(基底层分别为85.71%和71.43%;基底上层分别为76.19%和61.90%)。治疗后28天表皮全层PMLmRNA及强白的表达明显下降(基底层分别为9.52%和4.76%;基底上层分别为4.76%和4.76%)。结论:阿维A通过下调寻常型银屑病表皮中PML基因的表达抑制表皮角质形成细胞的增殖,并诱导其凋亡。
Objective: To determine the impact of acitretin on the expression of PML (Promyelocytic leukemia) gene and its protein in the epidemfis of the patients with psoriasis vulgaris. Methods: The expression of PML mRNA and its protein was detected by hybridization in situ and immunohistochemistry in the skin lesions of the patients with psoriasis vt~tris before and after the treatment with acitretin. Re~ts: There was high expression of PML mR.NA and its protein in the epidermis of the patients (basal layer: 85.71% and 71.43%, respectively; supra- basal layers: 76.19% and 61.90%, respectively) before the treatment of acitretin. The expression of PML mRNA and its protein was reduced markedly after 28 days of the treatment with acitretin (basal layer: 9.52% and 4.76%, re- spectively; supra- basal layers: 4.76% and 4.76%, respectively). Cevc.hision: Acitretin can inhibit proliferation and induce apoptosis of keratinocytes through down- regulating expression of PML gene in psoriatic epidermis.
