目的观察孕鼠围产期铁缺乏对仔鼠肝组织促红细胞生成素(EPO)水平的影响。方法将6周龄雌性健康Wistar大鼠随机分为两组,实验组饲低铁饲料,对照组饲普通饲料。2周后按2∶1比例与健康成年Wistar雄性大鼠交配,孕期继续各组饲料喂养直至分娩。孕末期检测孕鼠血红蛋白(Hb),平均红细胞容积(MCV),平均血红蛋白含量(MCH),红细胞分布宽度(RDW),铁蛋白(Fn),EPO;待孕鼠产仔,观测仔鼠的病死率和体质量;取仔鼠肝脏,制备组织匀浆,检测其胰岛素样生长因子-1(IGF-1)和EPO水平。结果与对照组比较,实验组孕鼠血清MCV、MCH、Fn水平降低(P均〈0.05),仔鼠每窝个数及肝组织IGF-1水平降低(P均〈0.05);与对照组比较,实验组孕鼠血清和仔鼠肝组织EPO水平升高,但无统计学差异。结论孕鼠围产期铁缺乏,不仅刺激孕鼠EPO分泌,同时也使仔鼠缺氧刺激EPO水平上升。
Objective To observe the effect of perinatal iron deficiency on the level of erythropoietin (EPO) in hepat- ic tissue of rat offspring. Methods Six-week-old healthy female Wistar rats were randomly divided into two groups : the experimental group and the control group. The rats in the experimental group and control group were fed on low iron or nor- mal diet respectively. After two weeks, according to the 2:1 ratio, the rats were mated with healthy adult male rats and continued to feed each diet until delivery. The levels of hemoglobin (Hb), mean corpuscular volume (MVC), mean cor- puscular hemoglobin (MCH) , red blood cell distribution width (RDW), ferritin (Fn) and EPO were detected. After the offspring were born, the mortality rate and weight were observed, the liver of newborn rats were collected for tissue homoge- nate to measure the levels of insulin-like growth factor-1 and EPO. Results Compared with the control group, the levels of MCV, MCH and Fn were decreased in the experimental group during perinatal period ( P 〈 0.05 ) and the number of per fitter and IGF-1 in tissue homogenate were lower (P 〈 0.05 ) ; in experimental group, the level of EPO in the pregnant rat serum and newborn rat hepatic tissue was increased compared with the control group, but there was no statistical difference. Conclusion Perinatal iron deficiency not only stimulates the secretion of EPO in the pregnant rats, but also make newborn rats lack of oxygen which stimulates the level of EPO to rise.
