目的:探讨针刺对血管性痴呆(VD)模型大鼠脑组织梗死体积和海马CA1区神经元损伤的影响。方法:96只雄性Wistar大鼠随机分为假手术组、模型组、针刺组和非穴组,采用双侧颈总动脉结扎的方法造模,假手术只分离血管不结扎;术后3d给予针刺治疗,针刺组取"百会"、双侧"足三里",非穴组在髂前上棘上2cm穴处针刺,每日1次,共12次。治疗结束后取大鼠脑组织,采用红四氮唑染色(TTC)观察各组大鼠脑组织梗死体积变化,运用尼氏染色方法观察各组大鼠海马CA1区神经元损伤情况。结果:与假手术组比较,模型组脑组织梗死体积显著增加(P〈0.01)。与模型组比较,针刺组、非穴组梗死面积均明显减小(P〈0.01)。与非穴组相比较,针刺组梗死体积显著减少(P〈0.01)。与假手术组比较,模型组海马CA1区神经元细胞数量显著减少(P〈0.01)。与模型组比较,针刺组、非穴组神经元细胞数量明显增加(P〈0.01)。与非穴组比较,针刺组海马神经元数量显著增加(P〈0.01)。结论:针刺可显著降低VD大鼠脑组织梗死体积,减轻海马CA1区神经元损伤。
Objective: To explore the influence of acupuncture on volume of cerebral infarction and neuronic damage of rats with vascular dementia. Methods: Ninety-six Male Wistar rats were randomly divided into sham-operation group, model group, acupuncture group and non-acupuncture point group. The rat models of vascular dementia(VD) were established by ligaturing bilateral carotid artery, and rats in the sham-operation group were isolated only without vascular ligation. Three days after surgery, acupuncture was given. Rats in acupuncture group acupuncture on Baihui(GV20) and Zusanli(S36), and rats in non-acupuncture point group acupuncture on 2cm above the anterior superior iliac spine once a day, a total of 12 times. Rats were sacrificed at the end of the treatment, the infarct volume of the rats was observed by TTC, and the injury of hippocampal CA1 neurons was observed by Nissl staining. Results: Compared with the sham-operated group, the infarct volume of the brain tissue in the model group was significantly increased(P〈0.01). Compared with the model group, the infarct area of acupuncture group and non-acupoint group were significantly decreased( P〈0.01). Compared with the non-acupoint group, the infarct volume of the acupuncture group was significantly reduced(P〈0.01). Compared with the sham-operation group, the number of neurons in the hippocampal CA1 region of the model group was significantly decreased(P〈0.01). Compared with model group, the number of neuron cells in acupuncture group and non-acupoint group increased significantly(P〈0.01). Compared with non-acupoint group, the number of hippocampal neurons in acupuncture group increased significantly(P〈0.01). Conclusion: Acupuncture cansignificantly reduce the infarct volume of VD rats and reduce the damage of hippocampal CA1 neurons.
