目的采用HPLC法测定傣药雅解沙把中巴马汀、小檗碱的含量。方法色谱柱为TSK gel ODS-100V(250 mm×4.6mm,5μm),流动相为乙腈-0.4%磷酸溶液(32∶68),检测波长345 nm,流速l m L·min-1,柱温40℃。结果巴马汀、小檗碱的线性范围分别为5.160~82.60、2.260~36.20μg·m L-1,平均回收率分别为99.37%、97.15%,RSD分别为1.12%、2.51%;雅解沙把中巴马汀、小檗碱的含量分别约为1.49、0.50 mg·g-1。结论所用方法简便、灵敏、重复性好,可用于测定雅解沙把中的两种生物碱。
OBJECTIVE To establish an HPLC method for the determination of palmatine and berberine in Yajieshaba. METHODS The TSK gel ODS- 100V column (250 mm×4.6mm,5μm) was used and the mobile phase was acetonitrile -0.4% phosphate (32 : 68) with the detection wavelength of 345 nm, the flow rate of 1 mL· min - 1 and the column temperature of 40 ℃. RESULTS μmL for palmatme and 2. 260 - 36.20 μg. mL- 1 for berberine. This method showed a good linearity in the range of 5. 160- 82.60 -1 The average recoveries were 99.37 % (RSD = 1.12% ) and 97.15 % (RSD = 2.51% ) for palmatine and berberine, respectively. Palma- tine and berberine contents in Yajieshaba were approximately 1.49 mg· g-1 and 0.50 mg. g-1. CONCLUSION The method is sample, sensitive and repeatabe. Palmatine and berberine can be considered as major active substances of Yajieshaba.
