目的:探讨不同浓度雌激素对颏舌肌成肌细胞胞内钙离子浓度的影响。方法:体外培养大鼠颏舌肌成肌细胞,缓冲液孵育颏舌肌成肌细胞使其处于正常与微酸环境下,激光共聚焦显微镜观察不同浓度雌激素作用下成肌细胞胞内钙离子的变化。结果:正常环境下,不同浓度雌激素对颏舌肌成肌细胞胞内钙离子影响不大。在微酸环境下,雌激素能够迅速下调成肌细胞胞浆钙离子强度,并在低水平重新形成新的平衡。结论:在一定条件下,雌激素能够下调颏舌肌成肌细胞胞内钙离子水平,维持肌细胞活性,从而减少肌肉的疲劳与损伤。这可能是雌激素对OSAHS起保护作用的原因之一。
Objective: To investigate the effect of estrogen on regulating the level of intracellular calciumion of ge- nioglossus(GG) myoblast. Methods: GG myoblasts were incubated in normal and slightly acidic culture buffer, then we used laser confocal microscope to detect the changes of intracellular calciumion in myoblast, which were treated with different concentrations of estrogen. Results: Under normal environment, estrogen didn't influence the intracel lular calcium of GG myoblast. However, the intracellular calcium fluorescent signal was decreased significantly via estrogen treated in slightly acidic state. Then the concentration of intracellular calcium reformed a new balance in the lower level. Conclusion: Physiological estrogen could quickly downregulate the concentration of intracellular cal- cium in slightly acidic state. It is good for maintaining the celluar activity which attenuates the damage and fatigue of the muscle. Our results may provide one of the reasons that estrogen could prevent the OSAHS.
