目的对缺血再灌注大鼠脑皮质区域超氧阴离子的产生进行可视化检测。方法用MitoSOXTM(超氧阴离子荧光标记探针)标记大鼠脑表区域神经细胞,共焦点激光显微镜活体观察在缺血(10min)再灌注(30min)过程中,脑表区域O2-产生的情况。结果缺血开始3min后,O2-产生开始增加,并一直持续到再灌注后10min停止增长。结论在缺血期及再灌注早期O2-生成增加,且动脉附近区域O2-产生量要多于其他区域。
Objective We examined reactive oxygen species (ROS) generation on cerebral ischemia/reperfusion rats by intravital fluorescence imaging. Methods In anesthetized adult rats, MitoSOX (a fluorescent dye for superoxide radical) was injected into cortices by a pressurized bolus. Through a closed cranial window,fluorescent images were taken with a confocal microscope on 10-minute forebrain ischemia and 30-min reperfusion. Results In isehemia and the early period of reperfusion, the MitoSOX fluorescence intensity significantly increased to 183%,and these increases were significant in the areas adjacent to the arteries. Conclusion O2- production increased in ischemia and the early period of reperfusion period. The O2- production was location-selective, being significant in the areas adjacent to the arteries. This method was useful for investigating intraeellular in situ ROS production.
