甘蓝型油菜自交不亲和临保性及其连锁AFLP标记

甘蓝型油菜自交不亲和临保材料在杂种优势利用和芸薹属自交不亲和性的机理研究方面具有重要的价值。以自交不亲和系S11300为受体、临保系97-wen238为供体,通过不亲和植株与供体连续回交构建BC6、BG和BC8群体对临保性的遗传进行了初步的分析,并检出与其连锁的分子标记。S11300×97-wen238F1的平均亲和指数小于1,表明97-wen238为甘蓝型油菜自交不亲和临保系。BC6、BC7和BC8群体的亲和指数集中分布在小于2和大于10两端,以亲和指数小于2为不亲和、大于10为亲和,3个回交世代不亲和植株与亲和植株的分离比均符合1:1,说明97-wen238的临保性受1对基因控制。以包含140个体的BG为作图群体,利用集团选择法,通过筛选1280对AFLP引物组合,得到了与临保性两侧连锁的3个AFLP标记MC07/P002、MC14/P004和EA12/MG07,遗传距离分别为7.8cM、11.7cM和6.6cM,为标记辅助选择自交不亲和临保系奠定了基础。

英文摘要:

Self-incompatibility (SI) temporary maintainer in Brassica napus plays an important role in self-incompatibility hybrid production and in detecting genetic and molecular bases of Brassica self-incompatibility. In this report, temporary maintainer was genetically analyzed using BC6, BC7, and BC8 generations derived from continuous back-crossing SI plants from combination SI1300 × 97-wen238 with SI temporary maintainer 97-wen238 as donor. AFLP markers linked to temporary maintainer were detected using BC7 generation. Average self-compatibility index (SCI) of SI1300 × 97-wen238 Fl was lower than one, showing that 97-wen238 could well maintain the self-incompatibility. SCI in each of BC6, BC7, and BCs was distributed in two distinct groups, less than two or more than ten. By taking plants whose SCI was less than two as self-incompatible and more than ten as self-compatible, a goodness-of-fit to 1:1 for self-incompatible plants to self- compatible plants in each of the BC6, BC7, and BCs generations was observed, suggesting that one gene controlled self- compatibility in SI maintainer. From markers amplified by 1280 AFLP primer pairs, three flanking markers, MC14/PO04, MC07/PO02 and EA12/MG07 were searched. Their length were about 480 bp, 320 bp, and 250 bp, and their genetic distance to SI locus were 7.8 cM, 11.7 cM, and 6.6 cM, respectively. These markers would be helpful for marker- assistant selection for temporary maintainer in rapeseed self-incompatibility hybrid breeding programs. Temporary maintainer and its use potentials in rapeseed and nuaker-assistant selection for temporary maintainer were discussed.
