目的通过蛋白组学技术分离并鉴定子宫内膜异位症(EMT)血瘀证、气滞血瘀证、肾虚血瘀证患者在位内膜的差异蛋白质,寻找EMT证候特异性蛋白质。方法分别取6例子宫内膜异位症血瘀证、气滞血瘀证、肾虚血瘀证患者增生期在位内膜组织和6例增生期正常子宫内膜组织标本,配对分为6组。提取组织总蛋白,通过双向电泳技术分离总蛋白,并得到6组双向电泳凝胶图谱。通过PDQuest7.1.1软件对图像进行分析,并检测出表达差异点。通过基质辅助激光解吸电离飞行时间质谱(MALDI—TOF—TOF/MS)及蛋白质数据库检索进行蛋白质鉴定。结果根据蛋白质的不同等电点和分子量,通过双向凝胶电泳和质谱分析技术可以分离人EMT患者及健康妇女在位内膜,发现EMT血瘀证患者与健康妇女在位内膜之间有差异蛋白表达,这些差异蛋白可能成为EMT发病的候选生物标志物。结论EMT血瘀证、气滞血瘀证、肾虚血瘀证患者在位内膜之间蛋白质组成分存在差异,这些差异可能是证候实质的内在根源。
OBJECTIVE To seperate EMT syndromes specific protein by isolating and identifying differences within eutopic protein in patients with EMT blood stasis syndrome, blood stasis due to qi stagnation, blood stasis due to kidney deficiency. METHODS The globle proteins of 6 endometrium tissue specimens of blood stasis syndrome in hyperplasia eutopic endometriosis and the normal endometrium were extracted and divided into 6 couples. Total protein tissue was extracted, which was separated by two-dimensional electrophoresis to attain 6 groups DE gel images. The image was analyzed through PDQuest7.1.1 and expression differences were detected. MALDI-TOF-TOF/MS was resoluted by matrix-assisted laser and protein was iden- tified in protein database. RESULTS (1)According to isoelectric point and molecular weight of proteins, the differential expressed endometriosis-associated protein of blood stasis syndrome and health women were separated and identified. And there were different expression of eutopic endometrium of three types of blood stasis syndrome, which might be the candidate bio- marker of endometriosis. CONCLUSION There are differences in protein constituents in patients with EMT blood stasis syndrome, blood stasis due to qi stagnation, blood stasis due to kidney deficiency, which may be the underlying reasons for syndrome.
