目的研究丹参酮ⅡA(TanIIA)对HL-60细胞的人端粒酶反转录酶(hTERT)表达的影响,探讨TanlI诱导肿瘤细胞凋亡的分子机制。方法用RPMI1640培养HL-60细胞,培养24h后分为3组['FanllA组、全反式维A酸(ATRA)组、对照组],分别加入500I.zg·L“TanⅡA、500Ixg·L。ATRA和0.1mL·L“二甲基亚砜治疗5d。加药前和加药后每天分别收集细胞,采用锥虫蓝染色后计数活细胞,流式细胞仪(FCM)检测ttL-60细胞凋亡率,半定量RT—PCR法测定HL-60细胞hTERT相对表达水平。结果与对照组比较,药物治疗2d后,TanlIA组和ATRA组HL-60细胞的生长均明显受到抑制(Pn〈0.05),rranⅡA组和ATRA组的抑制作用比较差异无统计学意义(P〉0.05)。TanllA组和ATRA组在药物治疗2d后细胞凋亡率分别为32.11%,23.31%,均明显高于对照组(6.08%)(P。〈0.05),且细胞凋亡率逐日上升,而对照组细胞凋亡率无明显变化。药物处理1d后,TanlIA组和ATRA组的HL-60细胞hTERT相对表达水平均明显低于对照组(Pn〈0.05),且表达水平逐日下降,而对照组表达水平无明显变化。结论丹参酮ⅡA抑制HL-60细胞生长、诱导细胞凋亡可能与下调hTERT基因表达水平有关。
Objective To investigate the effects of Tanshinone 11 A(Tan II A) on human telomerase reverse transcriptase (hTERT) ex- pression of human myeloid leukemia cell lines HL - 60, and to explore the molecular mechanisms underlying Tan II A - induced cancer cell apoptosis. Methods HL - 60 cells were cultured in RPMI 1640. After 24 hours, the cultured cells were divided into 3 groups [ Tan II A, all - trans retinoic acid(ATRA) and control group], and 500 μg .L-1 Tan Ⅱ A, 500 p,g Ⅱ L-1 ATRA and 0.1 mL . L-1 dimethyl sulfoxide were infused in different wells, respectively. The incubation was continued for another 5 days, and the cells were harvested and extracted for analy- sis each day. Trypan blue ceils assays were applied to measure the effects of Tan II A on the cell viability, and the changes in cell morphology were observed by microscope. Cell apoptosis was measured by flow cytometry. Semi - quantitative reverse transcriptase - polymerase chain reaction ( RT - PCR) was used to detect hTERT expression. Results The results showed that HL - 60 cell growth were inhibited significantly after 2 days of intervention with Tan lI A or ATRA compared with the control group ( P~ 〈 0.05 ), but there was no statistically significant differences between Tan 1] A and ATRA group ( P 〉 0.05 ). After 2 days of Tan 1I A or ATRA treatment, HL - 60 cell apoptosis was observed, and the apoptosis rates were 32.1 I% and 23.31% , respectively, which were significantly higher than 6.08% in the control group (P 〈 0.05 ). At the same time, cell apoptosis rates increased in Tan ]I A or ATRA group day by day, but it had no change in control group. After 1 day of Tan Ⅱ A or ATRA treatment, hTERT mRNA expression levels of HL - 60 cells were significantly lower than those of the control group (P 〈 0.05 ) , and downward trends were observed day by day, but hTERT mRNA expression levels had no change in the control group.
