目的比较65%乙醇和无水乙醇回流提取法从甜瓜蒂中提取葫芦素的效率;同时比较甜瓜蒂纤维和甜瓜蒂粉末中葫芦素含量的差异。方法分别采用65%乙醇和无水乙醇加热回流法制备甜瓜蒂粉末提取液;采用无水乙醇加热回流法制备甜瓜蒂纤维提取液。通过HPLC测定提取液中葫芦素B、E和I的浓度。结果甜瓜蒂粉末采用无水乙醇制备的提取液中葫芦素B、E浓度均高于65%乙醇制备的提取液中葫芦素B、E浓度,差异有统计学意义(P〈0.05);但葫芦素I浓度比较,差异无统计学意义(P〉0.05)。甜瓜蒂纤维采用无水乙醇制备的提取液中也含有较高浓度的葫芦素B、E、I。结论采用乙醇加热回流法从甜瓜蒂中提取葫芦素,无水乙醇提取效率高于65%乙醇。甜瓜蒂粉末中葫芦素含量虽高于甜瓜蒂纤维,但甜瓜蒂纤维中仍有较高含量的葫芦素,不应当作废弃物处理。
Objective To study and compare the extraction process of cucurbitacin from dry Cucumis melo pedicle by the heating reflux method with 65% ethanol and anhydrous ethanol and compare the cucurbitacin content of fibers and powders of Cucumis melo pedicle.Methods The powder extraction solution of Cucumis melo pedicle was prepared by the heating reflux method with 65% ethanol and anhydrous ethanol,and the fiber extraction solution was prepared by the heating reflux method with anhydrous ethanol. High Performance Liquid Chromatography(HPLC)method was adopted to assay the concentration of cucurbitacin B,E and I in the extraction solution.Results The concentration of cucurbitacin B and E in the extraction solution obtained by anhydrous ethanol was significantly higher than in the extraction solution obtained by 65% ethanol(P〈0.05),but there was no significant difference in the concentration of cucurbitacin I between the two groups(P〉0.05).The fibers of Cucumis melo pedicle contained the relatively high concentrations of cucurbitacin B,E and I.Conclusion Anhydrous ethanol is more effective in the heating reflux method than 65% ethanol in extracting the cucurbitacin.The content of cucurbitacin in powders of Cucumis melo pedicle is much higher than in its fibers,but the fibers should not be discarded as waste since it contains the relatively high concentration of cucurbitacin.
