目的通过生物信息学,预测菲律宾蛤仔原肌球蛋白的线性表位。方法对菲律宾蛤仔原肌球蛋白同源性、抗原性指数进行分析,利用在线工具预测及综合分析预测表位,进一步与人和猪原肌球蛋白序列对比初步筛选过敏原线性表位。结果 4种软体类的原肌球蛋白同源性较高,且原肌球蛋白的整体性抗原指数较高。菲律宾蛤仔原肌球蛋白中抗原表位区域较多,在与人、猪的序列比较后,最终预测出4个抗原表位。结论预测出了4个线性表位分别为Peptide 2 TLLQKKYTNLENEFDQVNEK,Peptide 6 EKQVKDAKYVAEEADRKYDE,Peptide 9 KDAENRAAEAERVVNKLQ和Peptide 10 ELLAEKEK YKAISDELDQ。
Objective To predict the linear epitopes of tropomyosin from Philippines clam by bioinformatics.Methods Homology,antigenic index analysis,prediction of table online tool and comprehensive analysis of the Philippines clam tropomyosin were detected,and were further compared with human and pig tropomyosin sequences to make preliminary screening of allergen linear epitope.Results Four kinds of tropomyosins from mollusks were highly homologous,and the overall antigen index of the tropomyosin was high.There were many epitopes in the tropomyosin of Philippines clam,and finally 4 epitopes were predicted after compared with human and pig tropomyosin sequences.Conclusion This study predicted 4 epitopes as followed:Peptide 2 TLLQKKYTNLENEF DQVNEK,Peptide 6 EKQVKDAKYVAEEADRKYDE,Peptide 9 KDAENRA AEAERVVNKLQ and Peptide 10 ELLAEKEKYKAISDELDQ.
