目的 研究用二氯化钴(CoCl2)模拟胃癌细胞缺氧微环境的合适浓度,探讨内皮细胞特异性分子-1(ESM-1)在胃腺癌SGC-7901/BGC-823细胞缺氧环境中的表达情况。方法以两种人胃腺癌细胞(SGC-7901/BGC-823)为研究对象,分别用不同浓度二氯化钴(CoCl2)处理人胃腺癌细胞后,用四唑化合物(MTS)法检测CoCl2对SGC-7901和BGC-823细胞增殖的影响;Westernblot分析不同浓度CoCl2处理的人胃腺癌SGC-7901和BGC-823细胞中低氧诱导因子-1α(HIF-1α)和ESM-1表达的变化。结果 ≤0.6mmol/L浓度的CoCl2对胃腺癌SGC-7901和BGC-823细胞的增殖和细胞活力没有明显影响。在0.5、0.6mmol/LCoCl2浓度时,HIF-1α蛋白表达明显增加,但ESM-1表达明显下降。结论 0.5、0.6mmol/LCoCl2可以模拟SGC-7901/BGC-823细胞的缺氧环境,在CoCl20.5、0.6mmol/L模拟细胞缺氧环境中,ESM-1的表达量与HIF-1α呈负相关。
Objective To investigate the effect of CoC12-induced chemical hypoxia on proliferation of human gastric carcinoma SGC-7901 and BGC-823 cells, and to study the expression of ESM-1 in the different degrees of hypoxia gastric carcinoma cells. Methods SGC-7901/BGC-823 cells were treated with 0. 1,0. 2,0. 3,0. 5,0. 6 mmol/L CoC12 for 24 hours. The expressions of HIF-ltx and ESM-1 were detected by Western blot. The change of cell prolif- eration was detected by MTS. Results Cobalt dichloride at the concentration lower than 0.6 mmol/L had no sig- nificant effect on proliferation of gastric carcinoma cell line SGC-7901 and BGC-823 cells. The Western blot analysis illustrated that the expression of ESM-1 declined obviously as the expression of HIF-1 ct increased. Conclusion 0. 5,0. 6 mmol/L Cobalt dichloride can induce cell hypoxia. ESM-1 is negatively correlated with HIF-1α.
