核糖体蛋白L11(ribosomal protein L11,RPL11)是核糖体60S大亚基的重要组成部分,对于核糖体的组装和合成是必须的。除此之外,研究发现真核生物中的RPL11还参与细胞周期阻滞、细胞凋亡及肿瘤发生等过程的调控。与抑癌蛋白ARF功能相类似,它能够与癌蛋白HDM2相互作用,激活P53抑痛蛋白的活性;与癌蛋白c-MYc相互作用,负反馈调控c—Myc的转录活性。推测RPL11本身可能具有肿瘤抑制功能。本文就RPL11参与调控HDM2-p53和c-MYC这两个与肿瘤密切相关的信号通路的研究进展进行综述。
Ribosomal protein L11 (RPL11) is a component of the large 60S ribosomal subunit which is essential for ribosomal biogenesis in eukaryotes. Ercept its classic role in ribosomal biogenesis, RPL11 also plays an important role in various other cellular processes, including cell cycle arrest, apoptosis and tumorigenesis. Like ARF, RPL11 can bind to HDM2 oncoprotein and suppresses HDM2-mediated p53 inhibition, leading to p53 activation. It also interacts with c-Myc oncoprotein and inhibits its transactivation activity. It is speculated that RPL11 itself may possess tumor suppressor function. This paper summarizes the recent findings of RPL11 in regulating the HDM2-p53 feedback loop and the c-Myc pathway.
