目的研究不同浓度顺铂对骨肉瘤143B细胞Notch通路的影响。方法不同浓度顺铂(2μM、4μM、6μM、8μM、10μM)处理不同时间(1 d、2 d、3 d、4 d、5 d))后,细胞增殖与活性检测(CCK-8法)测定顺铂对骨肉瘤细胞增殖抑制率的影响;实时定量PCR(qRT-PCR)分析Notch目的基因的相对表达量;Western blot及免疫荧光法半定量分析Notch目的基因的相对表达量。结果顺铂给药后143B细胞增殖受抑制,尤其是当浓度大于4μM时;qRT-PCR、Western blot及免疫荧光结果显示当顺铂浓度较低时(≤4μM),Notch信号通路激活,而浓度较高时(≥6μM)时则受抑制。结论顺铂对骨肉瘤Notch信号通路的影响具有剂量依赖性和时间依赖性,且必须在处理48 h后才能发挥其效应。
Objective To explore the mechanisms of various concentrations of cisplatin influencing the osteosarcoma proliferation.Methods Different concentrations of DDP( 2 μM,4 μM,6 μM,8 μM,10 μM) were used to treat 143 B cells in different days( 1 d,2 d,3 d,4 d,5 d),cell growth inhibition and IC50 were determined through cell counting kit-8 assay.qRT-PCR,western blot and immunofluorescence were used to analyze the relative expression of Notch target gene.Results The proliferation of 143B-cell were obviously inhibited after exposed to DDP,especially,the concentration of DDP more than 4 μM,qRT-PCR,Western blot and Immunofluorescence demonstrated that low concentration of DDP activates Notch signaling pathway,however,high concentration of DDP inhibited it( concentration≤4 μM).Conclusion The effect of cisplatin was proved to be dose-dependent and time-dependent,and 2 days was essential to play its role.
