通过给小鼠灌胃高脂饮食,观察蒲公英水提物对小鼠高脂性脂肪肝的治疗作用。设立空白组,模型组,阳性组及蒲公英水提物高、中、低剂量组(10、5、2.5g/kg),给小鼠腹腔注射地塞米松的同时,灌胃高脂饮食建立小鼠急性脂肪肝模型,观察小鼠肝脏指数,肝脏病理组织学变化,测定血清中谷丙转氨酶(ALT)和谷草转氨酶(AST),肝组织中甘油三酯(TG)和总胆固醇(TC)含量。结果与模型组相比,蒲公英水提物高、中剂量组可以显著或极显著降低脂肪肝小鼠的肝脏指数(P〈0.05或P〈0.01),降低血清中ALT和AST活性(P〈0.05或P〈0.01),降低肝组织中TG和TC含量(P〈0.05或P〈0.01);病理组织学观察蒲公英水提物高、中剂量组肝组织脂肪变性明显减轻。结果表明,蒲公英水提物对高脂饮食引起的小鼠高脂性脂肪肝有一定的治疗作用。
The therapeutic effect of dandelion extract on hyperlipidemic fatty liver in mice was observed by giving high-fat diet. The mice were divided into control group, model group, positive group and dandelion extract high,middle and low dose groups (10, 5 and 2.5 g/kg). Acute fatty liver mouse model was produced by intraperitoneally injecting dexamethasone and feeding high fat diet. The liver index, the liver histological change,ALT and AST in serum,TG and TC in liver tissue were measured. Compared with model group,dandelion extract high and middle dose groups could significantly decrease liver index of mice (P〈 0.05 or P〈0.01), decrease ALT and AST levels of serum (P〈0.05 or P〈0.01) and TG and TC content of the liver tissue(P〈0.05 or P〈0.01). The hepatic steatosis in dandelion extract high and middle dose groups was significantly reduced by pathological examination. The results showed that dandelion extract has certain therapeutic effect on fatty liver caused by high-fat diet in mice.
