目的探讨Th17细胞与相关转导因子在银屑病中的表达及意义。方法采用ELISA法检测寻常性银屑病患者及对照组血清IL-17,IL-23和IL-6水平;收集寻常性银屑病患者皮损及正常人皮肤作为对照,用免疫组化法检测STAT3和ROR_γt的表达;通过蛋白印迹法测定STAT3和RORγt含量;实时荧光定量PCR法检测STAT3 mRNA和ROR_γtmRNA的表达。结果银屑病患者血清IL-17,IL-23和IL-6的表达水平明显高于对照组,银屑病患者皮损STAT3和ROR_γt阳性率及蛋白含量和STAT3mRNA的表达均高于对照组,差异均有统计学意义(P均〈0.01),ROR_γt mRNA的表达与对照组相比差异无统计学意义(P〉0.05)。结论 Th17细胞在患者血清与相关转导因子在皮损中表达上调,STAT3和ROR_γt可能参与诱导Th17细胞的产生,这可能是Th17细胞与相关转导因子参与银屑病发生和发展的机制之一。
Objective To Investigate expression of Th17 cells and transduction factor in psoriatic and its significance. Methods Serum level of IL-17 ,IL-23 ,IL-6 were detected by ELISA in unusual psoriasis patients and control group. Expression of STAT3, RORYt were detected by immunohistochemical assay, Western blot and Real-time PCR assay. Results Serum levels of IL-17,IL-23, IL-6 expression was significantly higher than that in the control group (P 〈 0.01 ). Expression of STAT3, RORYt were higher in psoriasis patients than that in the control group(P 〈 0.01 ). While the tmRNA level of mRORYshowed no statistically significant compared to control control (P 〉 0.05). Conclusion Thl7 cells in serum and related transduction factors are upregulated in skin lesions. STAT3 ,RORYt may be involved in the induction of Th17 ceils which were involved in mechanism of psoriasis and development.
