目的研究西洋参茎叶皂苷(PQS)对大鼠局灶性脑缺血后caspase-9表达的影响,并进一步探讨其机制。方法采用线栓法阻断大鼠一侧大脑中动脉(MCA),制备局灶性脑缺血模型,RT-PCR技术检测大脑皮质凋亡相关基因caspase-9 mRNA表达。结果西洋参茎叶皂苷能明显降低局灶性脑缺血大鼠脑组织凋亡相关基因caspase-9 mRNA表达。结论西洋参茎叶皂苷对大鼠局灶性脑缺血具有保护作用,其机理可能与西洋参茎叶皂苷抑制神经元细胞凋亡有关。
Objective To investigate the effects and the further mechanism of Panax quinquefolium saponins from stems and leaves (PQS) on caspase-9. Methods The rats with incomplete cerebral ischemia were induced with line occlusion in the middle cerebral artery (MCA). Caspase-9 mRNA in the cortical neuron was detected with reverse transcription polymerase chain reaction (RT-PCR). Results Panax quinquefolium saponins significantly reduced the expression of caspase-9 mRNA. Conclusion Panax quinquefolium saponins have protecting effect on cerebral ischemia,and the mechanism is related to inhibition of cell apoptosis.
