以不同生长阶段的菜青虫五龄幼虫、预蛹和蛹为材料,通过对3个不同生长阶段的菜青虫双向电泳图谱的比较找到18个差异较明显的蛋白质斑点,并通过质谱分析技术和数据库信息检索最终鉴定出8个蛋白,包括4个代谢相关蛋白(氧化还原酶、组氨酸乙酰转运酶、多胺氧化酶和支酸变位酶)、3个调控蛋白(锌指类蛋白、转座酶、与KNA甲基化酶有关的假想蛋白)和1个与结构E—钙粘附蛋白相似的蛋白,其余10个蛋白质功能皆不明确.本文的结果为进一步研究菜青虫发育相关蛋白奠定基础,同时也为研发以这些差异蛋白为作用靶点的新型生物农药提供理论依据.
Proteomics research is to understand quantitatively, dymamically and wholly the organism on the protein level through which the relatie informations about physiology and biochemistry of the cell and regulation can be gained. The proteins were extracted from fifth larval stage worms, prepupae and pupae, separated by the 2-DE and identified by PMF method. By MALDI-TOF-MS methods and database search, the results indicated that 18 differential protein blots were found and 8 of them were identified, which including metabolism rehtive proteins, regulating relative proteins and signal transductional relative proteins. They can lay the foundation of researching relative proteins of imported cabbage worm and help develop new bio-pesticides acting on these differential proteins in theory.
