近年来,针对肿瘤代谢的研究已经成为肿瘤研究的新热点,而对代谢过程中关键调控因子的研究更是肿瘤代谢研究的重中之重,转录辅激活因子过氧化物酶体增殖物激活受体1,辅激活因子la(peroxisomeproliferatorsactivatedreceptorgammaCO—activator1alpha,PGC-1α)便是其中之一。PGC-1α是线粒体生物合成的关键调控因子,其在多种代谢性疾病中扮演着重要的角色,是氧化代谢和合成代谢的关键节点,参与了肿瘤代谢的调节,可以从氧化代谢和合成代谢两个方面调控肿瘤细胞的存活、增殖和迁移。本文对PGC-1α的结构及其相关转录因子进行了综述,探讨PGC-1α通过代谢调节肿瘤发生和发展的机制,并重点阐述了PGC-1α在肿瘤代谢研究中的进展,以期为后续以PGC-1α为靶标的抗肿瘤代谢的基础研究及药物研发提供新的理论参考。
The research on tumor metabolism has become a new hotspot in recent years, of which the key regulatory molecules that mediating tumor metabolism are a priority for all. The transcriptional co- activator peroxisome proliferator-activated receptor gamma co- activator 1alpha (PGC-1α) is one of these key regulatory molecules. PGC-1α has been demonstrated to be a crucial regulatory molecule involved in a variety of metabolic diseases. It is also a key molecule in regulation of tumor metabolism, mediating oxidative metabolism and anabolism to participate in regulation of survival, proliferation and migration of tumor cells. This paper reviews the advances in research on structure of PGC-la and its related transcription molecules, and explores the mechanism of PGC-1α in regulation of occurrence and development of tumors, especiallyfocuses on advances in the role of PGC-1α in tumor metabolism, which may provide theoretical references for fundamental studies on antitumor metabolism based on PGC-1α target as well as the related drug development.
