探讨不同浓度氡对永生化人支气管上皮细胞(HBE)氧化损伤的影响。体外培养HBE细胞,在氡浓度一定的条件下以不同染毒时间(20000Bq/m2,10min、20rain、40min)染毒HBE细胞,检测氡对HBE细胞活性氧(ROS)、超氧化物歧化酶(SOD)和丙二醛(MDA)的含量的影响,同时通过单细胞凝胶电泳(SCGE)检测细胞DNA链断裂情况。结果表明:随染毒时间延长,MDA和ROS含量明显上升(p〈0.05),SOD活力明显下降(p〈0.05),SCGE研究发现,各组HBE细胞Olive尾距随染毒时间延长显著增加,且均呈现明显的剂量反应关系。氡可诱导HBE细胞产生大量自由基造成DNA损伤,从而对HBE细胞产生毒效应。
To study the effect of oxidative damage of HBE cells induced by radon exposure, HBE ceils were exposed to radon (20 000 Bq/m3) with different exposed time (10 min, 20 min and 40 min). The reactive oxygen spe- cies (ROS) level, the malondialdehyde (MDA) and the superoxi dismutase (SOD) content in HBE cells was detected respectively. Single cell gel electrophoresis (SCGE) was applied for quantitative analysis of DNA damage in HBE cells induced by radon exposure. Compared with the control group, the level of ROS and the content of MDA in- creased (p〈0.05) significantly, while the content of SOD was decreased (p〈0.05) significantly. The SCGE results showed that radon could significantly increase the Olive tail moment in DNA of HBE cells compared with the control group (p〈0.05). Exposure to radon could increase the oxidative damage of HBE cells.
