[目的]探讨基因表达与蛋白表达之间的关系。[方法]以高油酸油莱近等基因系出苗后7d、5~6叶期和越冬期的叶片、花瓣和授粉后20-35d的种子为材料,利用定量PCR方法对5个iTRAQ分析得到的差异蛋白对应基因在不同生育期表达规律进行研究。[结果]结果发现,5-6叶期和越冬期叶片中,除蛋白芸薹属植物丝氨酸羟甲基转移酶对应基因外,其余4个基因在高油酸油菜材料中的表达量均高于低油酸油菜中的表达量。在授粉后20~35d的种子中除蛋白芸薹属植物丝氨酸羟甲基转移酶对应基因外,其余4个基因在高油酸油菜材料中的表达量均低于低油酸油菜中表达量。iTRAQ分析得到的与脂肪酸合成相关差异蛋白对应基因Brmsica napus putative 3-ketoacyl—CoA reduetase 2 mRNA(gil28565601)在营养生长期上调表达,生殖生长期下调表达。[结论]研究结果为加快油菜育种进程提供了借鉴。
[Objective] The aim was to explore relationship of gene expression and protein expression. [Method] In this study, 7 d after emergence, 5-6 leaves stage and wintering period of leaves, petals and seeds 20-35 d after pollination were used as raw materials to find out the expression rules in different development period. The five corresponding genes of the differential proteins which derived from the iTRAQ analysis were studied by Real-time quantitative PCR. [Result] The results showed that: 5-6 leaves stage and wintering period of leaves, except Brassica rapa serine hydroxymethyltransferase 4 (LOC103863581) (gil297800886), the rest of the four genes expressed in a high oleic acid rapeseed materials were higher than in low expression of oleic acid rapeseed. Seeds 20-35 d after pollination, except Brassica rapa probable acetyl-CoA acetyl transferase, the rest of the four genes expressed in a high oleie acid rapeseed materials were lower than the low expression of oleic acid rapeseed, iTRAQ analysis, and the differences in protein corresponding to genes related to fatty acid synthesis Brassica napus putative 3-ketoacyl-CoA reductase 2 mRNA (gil28565601) raised expression in vegetative period, lower expression reproductive stage. [Conclusion] The results provided references for speeding up the breeding process of rapeseed.
