目的研究8周自主跑轮运动对小鼠空间记忆能力及海马内NCAM、SCF等相关基因表达的影响。方法12只5周龄雄性C57BL/6小鼠,随机分为运动组(E,n=6)和对照组(C,n=6)。E组进行8周自主跑轮运动,C组安静饲养。第8周利用Morris水迷宫测试其学习记忆能力。运动结束后断颈处死小鼠,取双侧海马。用RT-PCR检测小鼠海马内NCAM、SCF、VEGF、IGF-1的mRNA表达水平。结果 Morris水迷宫实验显示,E组小鼠学习记忆能力显著强于C组;RT-PCR结果显示,E组小鼠海马内NCAM mRNA(P〈0.05)、SCF mRNA(P〈0.05)、VEGF mRNA(P〈0.05)和IGF-1 mRNA(P〈0.01)的表达水平较C组显著上调。结论 8周自主跑轮运动上调VEGF mRNA、IGF-1 mRNA的表达,增加了NCAM mRNA、SCF mRNA表达,提高了小鼠空间学习记忆能力。
Objective To investigate the effect of 8- week voluntary wheel running on the spatial memory,and the gene expression of NCAM,SCF in mice hippocampus. Methods 12 5- week old C57 BL /6 male mice were randomly divided into exercise group( E,n = 6) and control group( C,n = 6). The exercise group mice took wheel running for 8 weeks while the control group took no exercise. At the 8th week,12 mice were subjected to the Morris water maze test to evaluate its learning- memory ability. After the exercise,all mice were killed to obtain the hippocampus. RT- PCR were used to detect the gene expression of NCAM,SCF,VEGF,IGF- 1. Results According to the Morris water maze test,the spatial memory of exercise group is significantly better control group. Voluntary wheel running raised the expression of NCAM mRNA( P 〈 0. 05),SCF mRNA( P 〈 0. 05),VEGF mRNA( P 〈 0. 05),IGF- 1 mRNA( P 〈 0. 01). Conclusions 8 weeks of voluntary wheel running raised the expression of VEGF mRNA,IGF- 1 mRNA,increased NCAM mRNA,SCF mRNA,and enhanced the ability of spatial memory of mice.
