目的:观察心肌培养液中可溶性细胞因子对体外骨髓间充质干细胞(MSCs)向心肌细胞诱导分化过程的影响;方法:10μmol/L 5氮胞苷(5-aza)诱导大鼠骨髓间充质干细胞向心肌细胞分化后,实验分为两组:5-aza诱导组常规培养基培养,5-aza诱导+心肌条件培养组以1∶1混合的心肌条件培养液培养,观察与比较两组细胞形态,免疫细胞化学法和western blot技术分析心肌特异蛋白表达。结果:5-aza组细胞排列紊乱,5-aza诱导+心肌条件培养组细胞呈集落样排列,可见肌管形成,自诱导2周开始两组均见肌钙蛋白I(troponin I)阳性细胞,而troponin I、结蛋白(desmin)、连接蛋白-43(connexin-43)在诱导2周开始表达,3、4周达高峰,各时间点两组蛋白表达没有明显差异;结论:心肌条件培养液对心肌细胞的体外分化过程没有促进作用,但是可以加强体外诱导生成的心肌样细胞之间的有机排列。
