目的:探讨β2肾上腺素受体(β2-AR)在皮肤创伤修复上皮间质化(EMT)过程中的作用机制及意义。方法:采用免疫印迹法(Westernblot)、逆转录一聚合酶链式反应(RT-PCR)、免疫荧光技术等实验方法,体外观察β2-AR激动剂(ISO)和抑制剂(ICI)对上皮细胞间质化标志物(a-平滑肌肌动蛋白、波形蛋白、snail)表达的影响及创面皮肤上皮化标志物(E-钙黏附素)表达的变化。结果:β2-AR可通过介导皮肤创面愈合的EMT过程,促进表皮细胞向间质表型转化,且运动和迁移能力在该过程中不断增强。结论:β2-AR是皮肤上皮间质化的关键分子,其介导的EMT过程是皮肤创面愈合的生物学基础。
Objective: To explore the mechanism and significance of β2-adrenergic receptor (β2-AR) in the procedure of epithelial mesenchymal transition (EMT) during wound healing and tissue regeneration. Methods: The influence of β2-AR agonist (ISO) and inhibitor (ICI) on the expression of markers of mesenchymal transition (a-smooth muscle actin, vimentin, snail) or marker of epithelization (E-cadherin) in epidermis cells during wound healing and tissue regeneration was observed using western blot, reverse transcription polymerase chain reaction (RT-PCR) and immunofluorescence. Results: The epithelial cells gained mesenchymal phenotypic feature, and got stronger ability in movement and migration. β2-AR was a critical molecule which mediated EMT process. Conelusions: β2-AR is a key molecule in the procedure of EMT, and β2-AR mediated EMT is a biological basis of skin wound healing and matrix remodeling.
