HPLC测定不同配比麻黄汤水煎液中的麻黄碱和伪麻黄碱

目的:研究麻黄桂枝配伍对小鼠自主活动和脑内氨基酸水平的影响。方法:选择自主活动情况稳定的小鼠随机分为空白对照组、桂枝组(0.45 g/kg)、麻黄组(0.75 g/kg)和麻桂配伍组(1.5 g/kg),记录各组给药20,60,90,180,240 min后小鼠的水平活动次数,处死后迅速采集脑组织,采用高效液相色谱-荧光检测法进行小鼠大脑皮层内氨基酸含量测定。结果:与空白对照组比较,麻黄组(0.75 g/kg)小鼠在180 min时水平活动次数显著增加,麻黄与桂枝药对间存在交互作用,麻桂配伍组(1.5 g/kg)的水平活动次数较麻黄组有降低趋势。实验建立的色谱条件对脑组织匀浆液中17种氨基酸分离较好,日内和日间精密度的RSD分别〈8.09%、8.70%,平均回收率均〉81.68%。与空白对照组相比,在给药90 min后,麻黄组(0.75 g/kg)脑内抑制性氨基酸类神经递质(GABA、Ala)、缬氨酸(Val)、酪氨酸(Tyr)水平均明显降低,桂枝组(0.45 g/kg)、麻桂配伍组(1.5 g/kg)脑内GABA、Ala、Val、Tyr水品较麻黄组明显升高。结论:麻黄对小鼠中枢具有兴奋作用,桂枝的配伍能拮抗其中枢兴奋作用,其机制可能与抑制麻黄对脑内氨基酸水平的改变有关。

英文摘要:

Objective: To study the effects of compatibility of Cinnamomi Ramulus and Herbal Ephedrae on locomotor activity and amino acid levels in the brain in mice. Methods: The mice with stable locomotor activity rhythm were randomly divided into three groups. Their crossing times were automatically recorded at different time after drug administration. Subsequently,mice were sacrificed and the brain tissues were then immediately separated. The supernatants were used to measure the amino acid levels in each mouse brain by the high performance liquid chromatography with fluorescence detection. Results: Compared with the control group,treatment with Herbal Ephedrae( 0. 75 g / kg) obviously increased the locomotor activity at 180 min after drug administration( P〈0. 05). Meanwhile,there was an interaction between Cinnamomi Ramulus and Herbal Ephedrae,and the mice treated with Cinnamomi Ramulus and Herbal Ephedrae( 1. 5 g / kg) showed lower locomotor activity when compared with the mice that only treated with Herbal Ephedrae. Furthermore,the chromatographic conditions established in present study had a good resolution. The within and between day variation coefficients were lower than 8. 09% and 8. 70%,respectively. The spiked recoveries were both greater than 81. 68%. Compared with the control mice,Herbal Ephedrae( 0. 75 g / kg) treatment could significantly drease the contents of inhibitory neurotransmitters( like GABA,Ala),Val,and Tyr at 90 min of drug administration. In contrast,elevated levels of these above-mentioned inhibitory neurotransmitters were obaserved in Cinnamomi Ramulus and Herbal Ephedrae( 1. 5 g / kg)treated mice compared with the Herbal Ephedrae treated mice( P〈0. 05). Conclusion: Herbal Ephedrae exhibits a central excitatory effect in mice. And this excitatory effect can be abolished by the combined use of Cinnamomi Ramulus,which may be attributed to its lifting effect on inhibitory neurotransmitters.
