目的观察瞬时受体电位M2(transient receptor potential melastatin type 2,TRPM2)在舌癌组织及舌鳞状细胞癌SCC9细胞株中的表达,同时观察不同浓度H2O2激活TRPM2通道后对舌鳞状细胞癌SCC9细胞株的影响。方法通过细胞免疫化学、Western-Blot和RT-PCR方法检测TRPM2在舌癌组织、SCC9细胞株及正常舌体组织中的表达情况;通过MTT、台盼蓝染色方法检测不同浓度的H2O2对SCC9细胞株增殖、存活率的影响。结果 TRPM2在舌鳞状细胞癌SCC9细胞株和舌癌组织中呈阳性表达,在正常舌组织中阴性表达;不同浓度的H2O2明显抑制SCC9细胞株的增殖(P〈0.05),降低了SCC9细胞株的存活率(P〈0.05),并呈剂量依赖关系。结论 TRPM2在舌鳞状细胞癌SCC9细胞株及舌癌组织中的高表达提示离子通道蛋白可能参与调节肿瘤的发生发展。
Objective To investigate the expression of transient receptor potential melastatin type 2(TRPM2)in the human tongue squamous cell carcinoma tissue and cell line SCC9.The effects of different concentrations of H2O2 on SCC9 after ion channel was activated were observed.Methods Immunocytochemistry,Western-blotting and reverse transcription polymerase chain reaction(RT-PCR) were used to detect the expression of TRPM2 in the human tongue squamous cell carcinoma tissues,cell line SCC9 and normal tongue tissues;MTT assay and trypan blue staining assay were used to study the effects of different concentrations of H2O2 on the proliferation and cell viability of SCC9 respectively.Results The positive expression of TRPM2 was observed in human tongue squamous cell carcinoma tissues and cell line SCC9,while the expression was negative in normal tongue tissues.The proliferation and cell viability of SCC9 were significantly inhibited by different concentrations of H2O2(P0.05)in a dose dependent manner.Conclusions The high expression of TRPM2 in human tongue squamous cell carcinoma tissue and cell line SCC9 indicated that ion channel proteins may be involved in regulation of tumor development.
