[目的]制备99mTc-标记的壳聚糖一乳糖酸复合物(99mTc-LA-CS),观察99mTc—LA-CS在正常裸鼠体内的生物分布及去唾液酸糖蛋白受体(ASGP—R)介导的肝细胞靶向作用.[方法]采用氯化亚锡还原法构建99mTc-LA.CS复合物,尾静脉注射给予裸鼠99mTc—LA-CS后分别在10min,60min,2h处死,取血液、肝脏、脾脏、胃、肠、肾脏、肺和肌肉等组织,进行99mTc-放射性检测,计算各个脏器每克组织的放射性摄取比率(%ID/g),并进行体内生物分布分析.[结果]99mTc-标记率高于93%,生物分布分析结果显示在裸鼠肝脏中有较高的摄取,在60min时,肝脏摄取率高于70%ID/g.[结论]放射性核素99mTc-标记的LA-CS在肝细胞中有特异性摄取,提示99mTc-LA-CS可作为ASGP—R介导的肝脏显像剂用于肝细胞成像.
OBJECTIVE To prepare the chitosan-lactose acid complexes (99mTc-LA-CS) labeled by 99mTC, and to investigate the biodistribution of 99 mTc-LA-CS in normal nude mice and the targeting effect of hepatocyte mediated by asialoglycoprotein receptor(ASGP-R). METHODS The 99 mTc-LA-CS complexes were constructed by the stannous chloride reduction method, and were injectied to the nude mice tail vein, and then the mice were sacrificed at 10 rain, 60 min and 2 h. It was collected that the blood, liver, spleen, stomach, intestine, kidney, lung and muscle tissues to be detected the 99mTc radiation capability, and the radioactive uptake rate of per gram of tissue from different organs (% ID/g) were calculated and the biodistribution in vivo was analyzed. RESULTS 9amTc labeling ratio was more than 93%, and the biodistribution resUlts showed that high uptake was in the liver in nude mice, and the liver uptake rate was more than 70% ID/g at 60 rain. CONCLUSION Radionuclide labeled 99mTc LA-CS has specific uptake in liver cells, and it is indicated that the 99mTc-LA-CS can be used for liver cell imaging as liver imaging agent mediated by ASGP-R.
