目的探讨毛囊周期中,β-catenin在毛囊黑素谱系细胞中的表达变化。方法采用β-gal染色、石蜡切片免疫组化、RT-PCR、Western blot等方法检测Dct-Lac-Z CD1小鼠毛囊的黑素谱系细胞随着毛囊的周期其细胞数量、分布的变化及β-catenin的表达在mRNA和蛋白水平表达量及部位的变化。结果 WB及RT-PCR结果表明在毛囊周期中,β-cate-nin的表达总量由高到低依次为生长早期(P4、P29)、生长中期(P8)、静止期(P23)、退化期(P18),数据经统计学单因素方差分析,各组间存在显著差异(P〈0.05)。免疫组化结果显示在黑素谱系细胞中,β-catenin强表达于活化的黑素母细胞、黑素干细胞和分化过程中的黑素细胞,弱表达于静息的黑素干细胞、成熟分化的黑素细胞。结论β-catenin的上调能促进毛囊黑素谱系细胞的分化,其周期性的表达变化可能是毛发周期性色素生成的基础。
Objective To investigate β-catenin expression changes in hair follicle melanocyte lineage cells during the hair cycle.Methods RT-PCR,Western blot analysis,β-gal staining,and immunohistochemistry with paraffin section were adopted to detect the amount and distribution changes of melanocyte lineage cells in Dct-Lac-Z CD1 mouse hair follicles,and to identify β-catenin expression and distribution at mRNA and protein levels.Results Western blotting and RT-PCR results exhibited the protein and mRNA expression levels of β-catenin in the hair cycle [early anagen(P4 and P29)medium anagen(P8)telogen(P23)catagen(p18)].Significant differences were found between β-catenin expression levels of the four phases(P0.05),using one-way analysis of variance.Immunohistochemistry results showed that among melanocyte lineage cells,β-catenin was strongly expressed in activated melanoblasts,melanocyte stem cells(MSCs),and melanocytes in differentiation,and was weakly expressed in resting MSCs and differentiated melanocytes.Conclusion Upregulation of β-catenin can promote melanocyte lineage cell differentiation in hair follicles.The periodic change of β-catenin expression may be the basis of periodic generation of hair pigments.
