利用硅胶和Sephadex LH20柱层析分离法,从茯神干燥粉末提取物中得到5种化合物,运用NMR和MS等波谱法分析鉴定5种化合物分别为麦角甾醇过氧化物、茯苓酸、5-(4-羟苯基)二氢呋喃-2(3H)-酮、B—D—Glu(3—1)α-D—Glu(由2个葡萄糖组成的二糖化舍物)、仅一亚麻酸。茯神复方镇痛作用结果显示,热板法中只有茯神复方的水提物和水溶性三萜成分,具有明显的镇痛效果;扭体法实验结果显示,茯神复方的水提物、水溶性三萜成分、粗多糖和苓灵神蜜膏能明显减少小鼠的扭体次数,抑制因冰醋酸诱导的局部疼痛,提高了小鼠的痛阈,水溶性三萜成分的剂量为0.44g·kg^-1时,药效最为显著。
Five compounds were isolated from dry powders of Poria cum Radix Pini by silica gel using Sephadex LH-20 chroma- tography. Their structures were elucidated by spectroscopic methods including NMR and MS. The 5 kinds identified compounds were ergosterol peroxide, pachymie acid, dihydro-5-(4-hydroxyphenyl)-2(3H)- Furanone, α-D-Glu(3-1 )ct-D-Glu and α-linolenic. The abirritation of the compound of Poria cure Radix Pini showed that only the waterextract from the compound of Poria curn Radix Pini and water-solubility triterpenoids had a demonstrable analgesic effect in the Hot-plate test. In writhing test, the water-extract from the compound of Poria cum Radix Pini, the water-solubility triterpenoids, crude polysaccharide and Ling-ling Shen Honey Paste had previously decreased the writhing times of mice, and it also restrained the local pain induced from glacial acetic acid and enhanced the mice's pain threshold. When the dosage of water-solubility triterpenoids was 0. 44g·kg^-1, the pharmacodynamic effects was most prominent.
